Tea tree vegetative storage and preservation

The direct field preservation of tea tree vegetative bodies requires a large amount of land and labor. Therefore, the long-term preservation techniques of tea tree roots, seedlings, seeds, and pollen were studied.

The roots were preserved by sterilizing the lignified roots before germination in spring, and were stored in plastic bags with sterilized semi-dry soil or sphagnum moss and placed in an environment of 1 to 5°C. After 7 years of storage, adventitious buds can reach a maximum of 70%. When stored at 1°C for 6 years, the roots can still be regenerated. Stored at low temperatures, the consumption of carbohydrates in root tissue can be maintained for approximately 12 years. Root survival can be judged by the amount of potassium dissolved in the root.

The preservation of seedlings can be 2 years old cutting seedlings in the early spring to place, sealed and stored at 5 °C, can be stored for 4 years, the survival rate of different species between 20% to 26%. After the preserved tea plants were taken out and planted, the second year could be used as a hybrid material, and the result rate was 63% to 70%. The longest shelf life is up to 9 years.

After 5 years of storage at -80°C, pollen still has a germination rate of 70%, and after 12 years, the germination rate is 60%. This makes it possible to hybridize varieties with different flowering stages.
